Background

Immunoglobulin G (IgG), is one of the most abundant proteins in serum with normal levels between 8-17 mg/mL in blood. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions; to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants. This product is reddish transparent liquid, containing 0.03% Proclin300.

## Introduction

The Goat anti-Pig IgG (Immune serum) is a versatile antibody that can be used in various applications such as ELISA, Western Blotting, and Double Immunodiffusion. This protocol provides a comprehensive guide for using this antibody in these validated applications.

## ELISA Protocol

Reagents or buffers: Goat anti-Pig IgG (Immune serum), Pig IgG, ELISA plates, blocking buffer, washing buffer, substrate, and stop solution.

Sample preparation: Coat ELISA plates with Pig IgG and block with blocking buffer.

Antibody incubation: Incubate the plates with Goat anti-Pig IgG (Immune serum) at a dilution of 1:2000-20000 for 1-2 hours at room temperature.

Washing or detection: Wash the plates with washing buffer and detect with substrate.

Technical notes: Optimize the antibody dilution and incubation time for best results.

## Western Blotting Protocol

Reagents or buffers: Goat anti-Pig IgG (Immune serum), Pig IgG, SDS-PAGE gel, transfer buffer, blocking buffer, washing buffer, and detection reagents.

Sample preparation: Separate Pig IgG by SDS-PAGE and transfer to a membrane.

Antibody incubation: Incubate the membrane with Goat anti-Pig IgG (Immune serum) at a dilution of 1:1000-20000 for 1-2 hours at room temperature.

Washing or detection: Wash the membrane with washing buffer and detect with detection reagents.

Technical notes: Use a suitable secondary antibody for detection and optimize the antibody dilution and incubation time for best results.

## Double Immunodiffusion Protocol

Reagents or buffers: Goat anti-Pig IgG (Immune serum), Pig IgG, agarose gel, and electrophoresis buffer.

Sample preparation: Prepare wells in an agarose gel and fill with Goat anti-Pig IgG (Immune serum) and Pig IgG.

Antibody incubation: Allow the samples to diffuse and react for 24-48 hours at room temperature.

Washing or detection: No washing or detection steps are required.

Technical notes: Optimize the antibody dilution and reaction time for best results.

Precautions

Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for best results.
