Lab-ready steps

The Donkey Anti-Human IgG/AP antibody is a versatile tool for detecting human IgG in various applications. Below are the protocols for each validated application.

## Western Blotting Protocol

Reagents or buffers: Tris-buffered saline with Tween 20 (TBST), skim milk, and a suitable AP substrate.

Sample preparation: Separate proteins by SDS-PAGE and transfer to a membrane.

Antibody incubation: Incubate the membrane with the Donkey Anti-Human IgG/AP antibody at a dilution of 1:1000-10000 in TBST with 5% skim milk for 1 hour at room temperature.

Washing or detection: Wash the membrane three times with TBST, then detect using a suitable AP substrate.

Technical notes: Optimize the blocking conditions and antibody dilution to minimize background.

## IHC-P Protocol

Reagents or buffers: Phosphate-buffered saline (PBS), Triton X-100, and a suitable AP substrate.

Sample preparation: Fix and embed tissue samples, then section and mount on slides.

Antibody incubation: Incubate the sections with the Donkey Anti-Human IgG/AP antibody at a dilution of 1:100-500 in PBS with 0.1% Triton X-100 for 1 hour at room temperature.

Washing or detection: Wash the sections three times with PBS, then detect using a suitable AP substrate.

Technical notes: Perform antigen retrieval if necessary, and optimize the antibody dilution to achieve the best signal-to-noise ratio.

## IHC-F Protocol

Reagents or buffers: PBS, Triton X-100, and a suitable fluorescence substrate.

Sample preparation: Fix and permeabilize cells or tissue samples, then mount on slides.

Antibody incubation: Incubate the samples with the Donkey Anti-Human IgG/AP antibody at a dilution of 1:100-1000 in PBS with 0.1% Triton X-100 for 1 hour at room temperature.

Washing or detection: Wash the samples three times with PBS, then detect using a suitable fluorescence substrate.

Technical notes: Optimize the fixation and permeabilization conditions to preserve the antigenicity of the target protein.

## ELISA Protocol

Reagents or buffers: Coating buffer, blocking buffer, and a suitable AP substrate.

Sample preparation: Coat the ELISA plate with the antigen, then block with a suitable blocking buffer.

Antibody incubation: Incubate the plate with the Donkey Anti-Human IgG/AP antibody at a dilution of 1:1000-10000 in the blocking buffer for 1 hour at room temperature.

Washing or detection: Wash the plate three times with PBS, then detect using a suitable AP substrate.

Technical notes: Optimize the coating and blocking conditions to minimize non-specific binding.

Precautions

Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilution and reaction conditions to achieve the best results.
