Lab-ready steps

The Mouse Anti-Bovine IgG/APC is a highly specific antibody for detecting bovine IgG in various applications.

## Immunofluorescence (IF) Protocol

For IF, the following reagents are required: phosphate-buffered saline (PBS), 4% paraformaldehyde, and a mounting medium with DAPI.

Sample preparation involves fixing cells or tissue sections with 4% paraformaldehyde for 15 minutes, followed by permeabilization with 0.1% Triton X-100 for 10 minutes.

The antibody should be diluted to 1:100-1000 in PBS and incubated with the sample for 1 hour at room temperature.

CABter washing with PBS, a secondary antibody conjugated to a fluorescent dye can be applied if necessary.

Technical notes: Use a negative control without primary antibody to assess background fluorescence.

## Flow Cytometry (FCM) Protocol

For FCM, the following reagents are required: PBS, 1% bovine serum albumin (BSA), and a fixative such as 1% paraformaldehyde.

Sample preparation involves washing cells with PBS and resuspending them in PBS with 1% BSA.

The antibody should be diluted to 1:100-1000 in PBS with 1% BSA and incubated with the cells for 30 minutes on ice.

CABter washing with PBS, cells can be fixed with 1% paraformaldehyde and analyzed by flow cytometry.

Technical notes: Use a negative control without primary antibody to set the gate for positive staining.

Both IF and FCM require optimization of antibody dilution and incubation times to achieve the best results.

**IF Protocol**

Use assay conditions validated for IF, apply Mouse Anti-Bovine IgG/APC at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: FCM=1:100-1000;

IF=1:100-1000.

**FCM Protocol**

Use assay conditions validated for FCM, apply Mouse Anti-Bovine IgG/APC at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: FCM=1:100-1000;

IF=1:100-1000.

Precautions

Store at 4°C, handle with care, include negative controls, and optimize antibody dilution and incubation times for best results.
