Background

Alpha-1,4-N-Acetylglucosaminyltransferase 4, also known as A4GNT is a 45 kDa glycoprotein, member of the glycosyltransferase 32 family of proteins. A4GNT catalyzes the transfer of N-acetylglucosamine (GlcNAc) to core 2 branched O-glycans. A4GNT is a single-pass type II membrane protein associated with the Golgi apparatus and contains the conserved DXD motif involved in catalytic activity. It is expressed in stomach and pancreas, as well as in gastric cancer cells. A4GNT is not expressed in peripheral blood cells, making it a putative biomarker for pancreatic cancer. A4GNT and Mucin 6 expression is upregulated in the gastric mucosa of H. pylori infected patients. Lumenal domain of human A4GNT shares 72% aa sequence identity with mouse and rat A4GNT.

Protocol / Instructions

Recommended workflow
Optimized handling and experimental guidance
Review the suggested sequence below before starting the assay to keep sample handling, incubation, and downstream interpretation aligned.
Lab-ready steps

The Anti-A4GNT Polyclonal Antibody is a rabbit-derived antibody raised against a recombinant protein of human A4GNT. This antibody is recommended for use in Immunohistochemistry-Paraffin sections (IHC-P) at a dilution of 1:50-200.

IHC-P Protocol

Reagents or buffers required include the Anti-A4GNT Polyclonal Antibody, paraffin-embedded tissue sections, xylene, ethanol, antigen retrieval buffer, and a detection system such as DAB or alkaline phosphatase.

Sample preparation involves deparaffinization of tissue sections using xylene and ethanol, followed by antigen retrieval using a suitable buffer.

Antibody incubation is performed by applying the Anti-A4GNT Polyclonal Antibody at the recommended dilution and incubating for a suitable duration, typically 1-2 hours at room temperature or overnight at 4°C.

Washing is done using a buffer such as PBS or TBS, and detection is achieved using a suitable detection system.

Technical notes: Optimize the antibody dilution and incubation time for best results. Use a suitable negative control to verify the specificity of the staining.

Precautions

Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for best results.
