The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization.
The ActivAbTMAnti-GH2 Polyclonal Antibody is a rabbit-derived antibody directed against the human growth hormone 2 (GH2) protein. This antibody has been validated for use in ELISA, IHC-P, and WB applications.
For ELISA, use the ActivAbTMAnti-GH2 Polyclonal Antibody at a dilution of 1:2000-5000. Reagents needed include a 96-well plate, antigen, blocking buffer, and a detection system such as HRP-conjugated secondary antibody. Sample preparation involves coating the antigen onto the plate, followed by blocking and incubation with the primary antibody. Washing and detection steps should be performed according to the detection system's instructions. Technical notes: ensure proper antigen coating and blocking to minimize non-specific binding.
For IHC-P, use the ActivAbTMAnti-GH2 Polyclonal Antibody at a dilution of 1:50-200. Reagents needed include paraffin-embedded tissue sections, antigen retrieval buffer, and a detection system such as DAB. Sample preparation involves deparaffinization and antigen retrieval, followed by incubation with the primary antibody. Washing and detection steps should be performed according to the detection system's instructions. Technical notes: optimize antigen retrieval conditions for best results.
For WB, use the ActivAbTMAnti-GH2 Polyclonal Antibody at a dilution of 1:500-2000. Reagents needed include sample lysates, SDS-PAGE gel, transfer membrane, and a detection system such as ECL. Sample preparation involves lysing cells and separating proteins by SDS-PAGE, followed by transfer onto a membrane. Incubation with the primary antibody should be performed in blocking buffer, followed by washing and detection steps according to the detection system's instructions. Technical notes: ensure proper protein transfer and blocking to minimize non-specific binding.
Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilutions for best results.
