Lab-ready steps

The Avidin/HRP antibody is a versatile tool for detecting avidin in various applications. The following protocols are recommended for each validated application.

## Western Blotting Protocol

Reagents or buffers: PBS, Tween-20, non-fat dry milk, and ECL substrate.

Sample preparation: Separate proteins by SDS-PAGE and transfer to a PVDF membrane.

Antibody incubation: Incubate the membrane with Avidin/HRP antibody at a dilution of 1:500-5000 in PBS with 0.1% Tween-20 and 5% non-fat dry milk for 1 hour at room temperature.

Washing or detection: Wash the membrane three times with PBS containing 0.1% Tween-20, and detect using ECL substrate.

Technical notes: Optimize the antibody dilution and incubation time for best results.

## IHC-P Protocol

Reagents or buffers: PBS, Triton X-100, and DAB substrate.

Sample preparation: Fix and embed tissue samples, then section and mount on slides.

Antibody incubation: Incubate the slides with Avidin/HRP antibody at a dilution of 1:400-800 in PBS with 0.1% Triton X-100 for 1 hour at room temperature.

Washing or detection: Wash the slides three times with PBS, and detect using DAB substrate.

Technical notes: Use a suitable antigen retrieval method if necessary.

## IHC-F Protocol

Reagents or buffers: PBS, Triton X-100, and fluorescent secondary antibody.

Sample preparation: Fix and permeabilize cells or tissue samples, then incubate with primary antibody.

Antibody incubation: Incubate the samples with Avidin/HRP antibody at a dilution of 1:400-800 in PBS with 0.1% Triton X-100 for 1 hour at room temperature.

Washing or detection: Wash the samples three times with PBS, and detect using a fluorescent secondary antibody.

Technical notes: Optimize the antibody dilution and incubation time for best results.

## ELISA Protocol

Reagents or buffers: PBS, Tween-20, and TMB substrate.

Sample preparation: Coat ELISA plates with avidin and block with non-fat dry milk.

Antibody incubation: Incubate the plates with Avidin/HRP antibody at a dilution of 1:500-5000 in PBS with 0.1% Tween-20 for 1 hour at room temperature.

Washing or detection: Wash the plates three times with PBS containing 0.1% Tween-20, and detect using TMB substrate.

Technical notes: Optimize the antibody dilution and incubation time for best results.

Precautions

Store at 2-8°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for best results.
