This gene encodes a member of the transforming growth factor beta (TGFB) family of cytokines, which are multifunctional peptides that regulate proliferation, differentiation, adhesion, migration, and other functions in many cell types. Many cells have TGFB receptors, and the protein positively and negatively regulates many other growth factors. The secreted protein is cleaved into a latency-associated peptide (LAP) and a mature TGFB1 peptide, and is found in either a latent form composed of a TGFB1 homodimer, a LAP homodimer, and a latent TGFB1-binding protein, or in an active form composed of a TGFB1 homodimer. The mature peptide may also form heterodimers with other TGFB family members. This gene is frequently upregulated in tumor cells, and mutations in this gene result in Camurati-Engelmann disease.
## Introduction
The Anti-2,3,TGF-β1 Monoclonal Antibody is a mouse-derived antibody that targets the 2,3,TGF-β1 protein in human and mouse samples. This protocol outlines the procedure for using this antibody in Flow Cytometry (FCM) applications.
### Reagents and Buffers
- Anti-2,3,TGF-β1 Monoclonal Antibody (5uL/Test)
- Phosphate-buffered saline (PBS)
- Fluorescence-activated cell sorting (FACS) buffer
- Cell fixation and permeabilization solutions (if required)
### Sample Preparation
Prepare cell suspensions according to standard protocols. For intracellular staining, fix and permeabilize cells using appropriate solutions.
### Antibody Incubation
Incubate cells with the Anti-2,3,TGF-β1 Monoclonal Antibody (5uL/Test) in FACS buffer for 30 minutes to 1 hour at 4°C.
### Washing and Detection
Wash cells with FACS buffer to remove unbound antibody. Detect bound antibody using a fluorescent secondary antibody or a directly conjugated primary antibody.
### Technical Notes
Optimize antibody dilution and incubation times as needed. Use appropriate controls, such as isotype-matched antibodies, to verify specificity.
Use assay conditions validated for FCM, apply Anti-2,3,TGF-β1 Monoclonal Antibody at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: FCM 5uL/Test..
Store at 2-8°C. Handle with care, and avoid repeated freeze-thaw cycles. Use appropriate controls and optimize antibody dilution for best results.
