The Donkey Anti-Rabbit IgG/Gold 15nm is a versatile antibody conjugate suitable for various immunocytochemical and immunogold silver staining techniques.
For IEM, reagents such as 0.1% glutaraldehyde and 4% paraformaldehyde are used for sample fixation. Samples are then incubated with the Donkey Anti-Rabbit IgG/Gold 15nm at a dilution of 1:20-200. CABter antibody incubation, samples are washed with PBS and post-fixed with osmium tetroxide. Technical notes: optimal results are obtained with a 1-hour incubation at room temperature.
IGS involves the use of silver enhancement reagents to amplify the gold signal. Sample preparation involves fixation with 4% paraformaldehyde, followed by incubation with the primary antibody and then the Donkey Anti-Rabbit IgG/Gold 15nm at 1:20-200 dilution. CABter washing, samples are treated with silver enhancement reagents. Technical notes: monitor the silver enhancement reaction to avoid over-enhancement.
