Background

The protein encoded by this intronless gene is a G-protein coupled receptor for serotonin (5-hydroxytryptamine). Ligand binding activates second messengers that inhibit the activity of adenylate cyclase and manage the release of serotonin, dopamine, and acetylcholine in the brain. The encoded protein may be involved in several neuropsychiatric disorders and therefore is often a target of antidepressant and other psychotherapeutic drugs.

Protocol / Instructions

Recommended workflow
Optimized handling and experimental guidance
Review the suggested sequence below before starting the assay to keep sample handling, incubation, and downstream interpretation aligned.
Lab-ready steps

The Anti-5HT1B Polyclonal Antibody is a rabbit-derived antibody that targets the 5HT1B protein, a G-protein coupled receptor for serotonin. This antibody has been validated for use in Western Blotting (WB) applications.

## Western Blotting Protocol
For Western Blotting, the recommended dilution of the Anti-5HT1B Polyclonal Antibody is 1:1000-3000. The following protocol is recommended:

Reagents or Buffers: RIPA buffer, protease inhibitors, SDS-PAGE gel, transfer buffer, blocking buffer, and ECL substrate.

Sample Preparation: Harvest cells or tissue samples and lyse in RIPA buffer with protease inhibitors. Separate proteins by SDS-PAGE and transfer to a PVDF membrane.

Antibody Incubation: Incubate the membrane with the Anti-5HT1B Polyclonal Antibody at the recommended dilution in blocking buffer for 1-2 hours at room temperature or overnight at 4°C.

Washing or Detection: Wash the membrane with TBST and detect using an ECL substrate.

Technical Notes: The optimal dilution and incubation time may vary depending on the specific application and sample type. It is recommended to optimize these conditions for each experiment.

Precautions

Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for each experiment.
