## Introduction
The Donkey Anti-Goat IgG/Cy3 is a secondary antibody used for detecting goat IgG primary antibodies. It is conjugated to the fluorescent dye Cy3, which has an excitation wavelength of 550 nm and an emission wavelength of 570 nm.
Reagents or buffers: phosphate-buffered saline (PBS), 4% paraformaldehyde, 0.1% Triton X-100, blocking buffer (e.g., 5% normal donkey serum).
Sample preparation: fix cells or tissue sections with 4% paraformaldehyde for 10-15 minutes, then permeabilize with 0.1% Triton X-100 for 10-15 minutes.
Antibody incubation: incubate samples with primary goat antibody, followed by incubation with Donkey Anti-Goat IgG/Cy3 at a dilution of 1:50-500 in blocking buffer for 1 hour at room temperature.
Washing or detection: wash samples three times with PBS, then visualize using a fluorescence microscope with a Cy3 filter set.
Technical notes: optimize the dilution of the Donkey Anti-Goat IgG/Cy3 and the incubation time to achieve the best signal-to-noise ratio.
Use assay conditions validated for IF, apply Donkey Anti-Goat IgG/Cy3 at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: IF=1:50-500.
Store at 4°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for best results.
