The Biotin/HRP antibody is a versatile tool for detecting biotinylated molecules in various applications. Below are the protocols for the validated applications.
Reagents or buffers: PBS, Tween-20, non-fat dry milk, biotinylated molecular weight markers.
Sample preparation: Separate proteins by SDS-PAGE and transfer to a PVDF membrane.
Antibody incubation: Incubate the membrane with the Biotin/HRP antibody at a dilution of 1:1000-10000 in PBS with 0.1% Tween-20 and 5% non-fat dry milk for 1 hour at room temperature.
Washing or detection: Wash the membrane three times with PBS containing 0.1% Tween-20, and detect using a chemiluminescent substrate.
Technical notes: Use biotinylated molecular weight markers as positive controls.
Reagents or buffers: PBS, Triton X-100, normal serum, DAB substrate.
Sample preparation: Fix and embed tissue samples, and cut into 4-6 micron sections.
Antibody incubation: Incubate the sections with the Biotin/HRP antibody at a dilution of 1:100-1000 in PBS with 0.1% Triton X-100 and 1% normal serum for 1 hour at room temperature.
Washing or detection: Wash the sections three times with PBS, and detect using a DAB substrate.
Technical notes: Use biotinylated antibodies as positive controls.
Reagents or buffers: PBS, Tween-20, non-fat dry milk, TMB substrate.
Sample preparation: Coat ELISA plates with biotinylated antigens.
Antibody incubation: Incubate the plates with the Biotin/HRP antibody at a dilution of 1:1000-10000 in PBS with 0.1% Tween-20 and 5% non-fat dry milk for 1 hour at room temperature.
Washing or detection: Wash the plates three times with PBS containing 0.1% Tween-20, and detect using a TMB substrate.
Technical notes: Use biotinylated antibodies as positive controls.
Store at 2-8°C, handle with care, use appropriate controls, and optimize antibody dilutions for each application.
