## Introduction

The Goat Anti-Guinea Pig IgG/Bio antibody is designed for various applications including Western Blotting (WB), Immunohistochemistry-Paraffin (IHC-P), Immunohistochemistry-Frozen (IHC-F), Immunofluorescence (IF), Flow Cytometry (FCM), and Enzyme-Linked Immunosorbent Assay (ELISA). This protocol outlines the procedures for each validated application.

## Western Blotting Protocol

For Western Blotting, dilute the antibody to 1:1000-10000 in the recommended dilution buffer. Prepare samples by lysing cells or tissues in an appropriate lysis buffer. Separate proteins by SDS-PAGE and transfer to a membrane. Block the membrane with a blocking buffer, then incubate with the diluted antibody. Wash the membrane with a washing buffer, and detect using a suitable detection system.

## IHC-P Protocol

For IHC-P, dilute the antibody to 1:100-500. Prepare paraffin-embedded tissue sections and deparaffinize them. Antigen retrieval may be necessary. Block the sections with a blocking buffer, then incubate with the diluted antibody. Wash with a washing buffer, and detect using a suitable detection system.

## IHC-F Protocol

For IHC-F, dilute the antibody to 1:100-1000. Prepare frozen tissue sections and fix them. Block the sections with a blocking buffer, then incubate with the diluted antibody. Wash with a washing buffer, and detect using a suitable detection system.

## Immunofluorescence Protocol

For IF, dilute the antibody to 1:100-1000. Prepare cell samples or tissue sections and fix them. Block the samples with a blocking buffer, then incubate with the diluted antibody. Wash with a washing buffer, and detect using a fluorescent secondary antibody.

## Flow Cytometry Protocol

For FCM, dilute the antibody to 1:100-1000. Prepare cell suspensions and fix them if necessary. Block the cells with a blocking buffer, then incubate with the diluted antibody. Wash with a washing buffer, and detect using a suitable detection system.

## ELISA Protocol

For ELISA, dilute the antibody to 1:1000-10000. Prepare antigen-coated plates and block them with a blocking buffer. Incubate with the diluted antibody, then wash with a washing buffer. Detect using a suitable detection system.

## Technical Notes

Optimize the dilution and incubation time for each application as needed. Use appropriate controls to ensure specificity and sensitivity.

**IF Protocol**

Use assay conditions validated for IF, apply Goat Anti-Guinea Pig IgG/Bio at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: WB=1:1000-10000;

IHC-P=1:100-500;

IHC-F=1:100-1000;

IF=1:100-1000;

ELISA=1:1000-10000;

FCM=1:100-1000.

**FCM Protocol**

Use assay conditions validated for FCM, apply Goat Anti-Guinea Pig IgG/Bio at the recommended working dilution, include proper positive and negative controls, perform thorough wash steps, and document incubation time, temperature, and detection conditions for reproducible performance. Recommended dilution: WB=1:1000-10000;

IHC-P=1:100-500;

IHC-F=1:100-1000;

IF=1:100-1000;

ELISA=1:1000-10000;

FCM=1:100-1000.

Precautions

Store at 2-8°C. Handle with care. Use appropriate controls and optimize conditions for each application to ensure specificity and sensitivity.
