Background

Immunoglobulin G (IgG), is one of the most abundant proteins in serum with normal levels between 8-17 mg/mL in blood. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions; to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants. This product is reddish transparent liquid, containing 0.03% Proclin300.

## Introduction

The Goat anti-Rat IgG (Immune serum) is a versatile antibody that can be used in various applications such as ELISA, Western Blotting, and Double Immunodiffusion. This protocol provides a comprehensive guide for using this antibody in these validated applications.

## ELISA Protocol

Reagents or buffers: Goat anti-Rat IgG (Immune serum), Rat IgG, ELISA plates, blocking buffer, washing buffer, substrate.

Sample preparation: Coat ELISA plates with Rat IgG and block with blocking buffer.

Antibody incubation: Incubate the plates with Goat anti-Rat IgG (Immune serum) at a dilution of 1:5000-50000 for 1-2 hours.

Washing or detection: Wash the plates with washing buffer and detect with substrate.

Technical notes: Optimize the dilution of the antibody and the incubation time for best results.

## Western Blotting Protocol

Reagents or buffers: Goat anti-Rat IgG (Immune serum), Rat IgG, SDS-PAGE gel, transfer buffer, blocking buffer, washing buffer, substrate.

Sample preparation: Separate Rat IgG by SDS-PAGE and transfer to a membrane.

Antibody incubation: Incubate the membrane with Goat anti-Rat IgG (Immune serum) at a dilution of 1:1000-20000 for 1-2 hours.

Washing or detection: Wash the membrane with washing buffer and detect with substrate.

Technical notes: Use a suitable blocking buffer to reduce non-specific binding.

## Double Immunodiffusion Protocol

Reagents or buffers: Goat anti-Rat IgG (Immune serum), Rat IgG, agarose gel, buffer.

Sample preparation: Prepare a gel plate with wells and add Rat IgG and Goat anti-Rat IgG (Immune serum) at a dilution of 1:4-32.

Antibody incubation: Allow the gel to incubate for several hours or overnight to allow the formation of precipitin lines.

Washing or detection: No washing or detection is required as the precipitin lines are visible.

Technical notes: Optimize the concentration of the antibody and the incubation time for best results.

Precautions

Store at 2-8°C, handle with care, use appropriate controls, and optimize antibody dilution and incubation time for best results.
