Background

The scaffolding protein encoded by this gene is the main component of the caveolae plasma membranes found in most cell types. The protein links integrin subunits to the tyrosine kinase FYN,an initiating step in coupling integrins to the Ras-ERK pathway and promoting cell cycle progression. The gene is a tumor suppressor gene candidate and a negative regulator of the Ras-p42/44 mitogen-activated kinase cascade. Caveolin 1 and caveolin 2 are located next to each other on chromosome 7 and express colocalizing proteins that form a stable hetero-oligomeric complex. Mutations in this gene have been associated with Berardinelli-Seip congenital lipodystrophy. Alternatively spliced transcripts encode alpha and beta isoforms of caveolin 1.

Lab-ready steps
Western Blotting Protocol

Reagents or buffers: RIPA buffer, protease inhibitor cocktail, SDS-PAGE gel, transfer buffer, blocking buffer, and ECL substrate.

Sample preparation: Lyse cells in RIPA buffer with protease inhibitors, then separate proteins by SDS-PAGE and transfer to a membrane.

Antibody incubation: Incubate the membrane with Anti-CAV1 Recombinant Antibody at a suitable dilution in blocking buffer for 1-2 hours at room temperature or overnight at 4°C.

Washing or detection: Wash the membrane with TBST, then incubate with a secondary antibody conjugated to HRP, and detect using ECL substrate.

Technical notes: Optimize the antibody dilution and incubation time for best results.

IHC-P Protocol

Reagents or buffers: Xylene, ethanol, antigen retrieval buffer, blocking buffer, and DAB substrate.
