Background

Immunoglobulin G (IgG), is one of the most abundant proteins in serum with normal levels between 8-17 mg/mL in blood. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions; to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants. This product is reddish transparent liquid, containing 0.03% Proclin300.

Protocol / Instructions

Recommended workflow
Optimized handling and experimental guidance
Review the suggested sequence below before starting the assay to keep sample handling, incubation, and downstream interpretation aligned.
Lab-ready steps
ELISA Protocol

Reagents or buffers: Goat anti-Mouse IgG, coating buffer, blocking buffer, sample diluent, substrate.

Sample preparation: Coat the plate with the antigen, block, and add samples.

Antibody incubation: Add the diluted Goat anti-Mouse IgG, incubate, and wash.

Washing or detection: Add substrate, measure absorbance.

Technical notes: Optimize the coating and blocking conditions.

Western Blotting Protocol

Reagents or buffers: Goat anti-Mouse IgG, transfer buffer, blocking buffer, wash buffer.

Sample preparation: Separate proteins by SDS-PAGE, transfer to membrane.

Antibody incubation: Block the membrane, add the diluted Goat anti-Mouse IgG, incubate, and wash.

Washing or detection: Detect using a suitable method, such as ECL.

Technical notes: Optimize the blocking and washing conditions.

Double Immunodiffusion Protocol

Reagents or buffers: Goat anti-Mouse IgG, agar, antigen.

Sample preparation: Prepare the agar plate, add the antigen and Goat anti-Mouse IgG.

Antibody incubation: Allow the diffusion to occur.

Washing or detection: Observe the precipitin lines.

Technical notes: Control the temperature and humidity during the diffusion process.

Precautions

Store at 2-8°C, handle with care, use appropriate controls, and optimize conditions for best results.
