This gene is a classical cadherin from the cadherin superfamily. The encoded protein is a calcium dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats,a transmembrane region and a highly conserved cytoplasmic tail. The protein functions during gastrulation and is required for establishment of left-right asymmetry. At certain central nervous system synapses, presynaptic to postsynaptic adhesion is mediated at least in part by this gene product.
The Anti-Cadherin 2 Recombinant Antibody is a valuable tool for detecting Cadherin 2 in various applications. Below are the protocols for the validated applications.
Reagents or buffers: RIPA buffer, SDS-PAGE gel, transfer buffer, blocking buffer, and ECL substrate.
Sample preparation: Lyse cells in RIPA buffer, separate proteins by SDS-PAGE, and transfer to a membrane.
Antibody incubation: Incubate the membrane with the Anti-Cadherin 2 Recombinant Antibody at a suitable dilution in blocking buffer for 1-2 hours at room temperature.
Washing or detection: Wash the membrane with TBST, incubate with a secondary antibody, and detect using ECL substrate.
Technical notes: The expected band size is approximately 100 kDa.
Reagents or buffers: Xylene, ethanol, antigen retrieval buffer, blocking buffer, and DAB substrate.
Sample preparation: Fix and embed tissue samples, section them, and deparaffinize with xylene and ethanol.
Antibody incubation: Incubate the sections with the Anti-Cadherin 2 Recombinant Antibody at a suitable dilution in blocking buffer for 1-2 hours at room temperature.
Washing or detection: Wash the sections with PBS, incubate with a secondary antibody, and detect using DAB substrate.
Technical notes: Antigen retrieval using heat-mediated epitope retrieval is recommended.
Reagents or buffers: PBS, FACS buffer, and a fluorescent secondary antibody.
Sample preparation: Prepare single-cell suspensions from tissues or cells, and fix them if necessary.
Antibody incubation: Incubate the cells with the Anti-Cadherin 2 Recombinant Antibody at a suitable dilution in FACS buffer for 30 minutes to 1 hour at 4°C.
Washing or detection: Wash the cells with FACS buffer, incubate with a fluorescent secondary antibody, and analyze by flow cytometry.
Technical notes: Use a suitable isotype control to verify the specificity of the staining.
Store at -20°C, handle with care, use appropriate controls, and optimize antibody dilutions for each application.
