Background

Monomeric IgA constitutes 5-15 % of the serum immunoglobulins whereas dimeric IgA is localized to mucosa surfaces such as saliva, gastrointestinal secretion, bronchial fluids and milk. Mucosal IgA plays a major role in host defence by neutralising infectious agents at mucosal surfaces. The production is usually local and antigen specific IgA producing B-cells can be found in regions under the lamina propria where they mature into dimeric IgA producing plasma cells. IgA deficiency is the most common immunodeficiency that may CABfect both serum and mucosal produced IgA.

Protocol / Instructions

Recommended workflow
Optimized handling and experimental guidance
Review the suggested sequence below before starting the assay to keep sample handling, incubation, and downstream interpretation aligned.
Lab-ready steps

The Goat anti-Human IgA (Immune serum) is designed for use in various immunological applications.

## Double Immunodiffusion Protocol

Double Immunodiffusion is a technique used to detect and quantify antigens and antibodies in a sample.

Reagents or buffers: 1% agarose in PBS, PBS, antigen and antibody samples.

Sample preparation: Prepare the antigen and antibody samples according to the manufacturer's instructions.

Antibody incubation: Add the antibody to the gel and incubate at room temperature for 24-48 hours.

Washing or detection: Observe the precipitation lines and record the results.

Technical notes: The recommended dilution for Double Immunodiffusion is 1:16-32.

Precautions for handling and optimization should be considered to ensure accurate results.

Precautions

Store at 2-8°C. Handle with care, avoid repeated freeze-thaw cycles. Use appropriate controls and optimize antibody dilution for best results.
