Background

This gene encodes the most abundant protein in human blood. This protein functions in the regulation of blood plasma colloid osmotic pressure and acts as a carrier protein for a wide range of endogenous molecules including hormones, fatty acids, and metabolites, as well as exogenous drugs. Additionally, this protein exhibits an esterase-like activity with broad substrate specificity. The encoded preproprotein is proteolytically processed to generate the mature protein. A peptide derived from this protein, EPI-X4, is an endogenous inhibitor of the CXCR4 chemokine receptor.

The Anti-ALB Polyclonal Antibody is a versatile tool for detecting ALB in various applications.

## ELISA Protocol

For ELISA, coat the plate with the antigen and block with 5% BSA in PBS. Incubate with the Anti-ALB Polyclonal Antibody at a dilution of 1:500-1000 in PBS with 1% BSA. Use a secondary antibody conjugated to HRP and detect with a chromogenic substrate.

## IF/ICC Protocol

For IF/ICC, fix cells with 4% PFA and permeabilize with 0.1% Triton X-100. Incubate with the Anti-ALB Polyclonal Antibody at a dilution of 1:50-200 in PBS with 1% BSA. Use a secondary antibody conjugated to a fluorescent dye and detect with a fluorescence microscope.

## WB Protocol

For WB, separate proteins by SDS-PAGE and transfer to a PVDF membrane. Block with 5% milk in TBS-T and incubate with the Anti-ALB Polyclonal Antibody at a dilution of 1:500-1000 in TBS-T with 1% milk. Use a secondary antibody conjugated to HRP and detect with a chemiluminescent substrate.

Technical notes: Optimize the antibody dilution and incubation time for each application to achieve the best results.

**Western Blotting Protocol**
  1. Prepare lysates in SDS-compatible lysis buffer containing protease inhibitors, clarify by centrifugation, and denature samples at 95–100°C for 5 minutes.
  2. Resolve proteins by SDS-PAGE and transfer to PVDF or nitrocellulose membrane.
  3. Block with 5% BSA or nonfat milk in TBST for 1 hour at room temperature.
  4. Incubate the membrane with Anti-ALB Polyclonal Antibody diluted in antibody dilution buffer overnight at 4°C with gentle agitation. Recommended dilution: WB 1:500-1000. IF/ICC 1:50-200..
  5. Wash 3 times in TBST, then incubate with species-appropriate HRP-conjugated secondary antibody for 1 hour at room temperature.
  6. Wash again, develop with ECL substrate, and document target band intensity together with molecular weight marker and exposure conditions.
