Background
Immunoglobulin G (IgG)，is one of the most abundant proteins in serum with normal levels between 8-17 mg/mL in blood. IgG is important for our defence against microorganisms and the molecules are produced by B lymphocytes as a part of our adaptive immune response. The IgG molecule has two separate functions;to bind to the pathogen that elicited the response and to recruit other cells and molecules to destroy the antigen. The variability of the IgG pool is generated by somatic recombination and the number of specificities in an individual at a given time point is estimated to be 1011 variants.
Protocol / Instructions
Reagents or buffers: Tris-buffered saline with Tween 20 (TBST), non-fat dry milk, Goat anti-Bovine IgG antibody.
Sample preparation: Prepare bovine IgG samples and load onto an SDS-PAGE gel. Transfer the separated proteins to a PVDF membrane.
Antibody incubation: Incubate the membrane with Goat anti-Bovine IgG antibody at a dilution of 1:1000-20000 in TBST with 5% non-fat dry milk for 1 hour at room temperature.
Washing or detection: Wash the membrane three times with TBST and detect using a suitable detection system.
Technical notes: Optimize the antibody dilution and incubation time for best results.
