This gene encodes a protein that is highly similar to mouse and rat kynurenine aminotransferase II. The rat protein is a homodimer with two transaminase activities. One activity is the transamination of alpha-aminoadipic acid, a final step in the saccaropine pathway which is the major pathway for L-lysine catabolism. The other activity involves the transamination of kynurenine to produce kynurenine acid, the precursor of kynurenic acid which has neuroprotective properties. Several transcript variants encoding two different isoforms have been found for this gene.
The Anti-AADAT Polyclonal Antibody is a rabbit-derived antibody that recognizes the AADAT protein. For optimal results, follow the recommended dilutions and protocols for each validated application.
Reagents or buffers: Tris-buffered saline with Tween 20 (TBST), skim milk, and ECL reagent.
Sample preparation: Prepare protein samples and separate them by SDS-PAGE. Transfer the proteins to a PVDF membrane.
Antibody incubation: Incubate the membrane with the Anti-AADAT Polyclonal Antibody at a dilution of 1:2000-5000 in TBST with 5% skim milk for 1-2 hours at room temperature.
Washing or detection: Wash the membrane three times with TBST, then detect the signal using ECL reagent.
Technical notes: The expected band size is approximately 54 kDa.
Reagents or buffers: Xylene, ethanol, antigen retrieval buffer, and DAB substrate.
Sample preparation: Prepare paraffin-embedded tissue sections and deparaffinize them using xylene and ethanol.
Antibody incubation: Incubate the sections with the Anti-AADAT Polyclonal Antibody at a dilution of 1:25-100 in antibody diluent for 1-2 hours at room temperature.
Washing or detection: Wash the sections with PBS, then detect the signal using DAB substrate.
Technical notes: Antigen retrieval using heat-mediated epitope retrieval is recommended.
Store at -20°C. Handle with care, and avoid repeated freeze-thaw cycles. Use appropriate controls and optimize antibody dilutions for each application.
