β-Actin is one of six different actin isoforms that have been identified. The actin molecules found in cells of various species and tissues tend to be very similar in their immunological and physical properties. Therefore,Antibodies against β-Actin are useful as loading controls for Western Blotting. However it should be noted that levels of β-Actin may not be stable in certain cells. For example,expression of β-Actin in adipose tissue is very low and therefore β-Actin should not be used as loading control for these tissues.
The Anti-ACTB Monoclonal Antibody is a mouse-derived antibody that recognizes human, mouse, and rat ACTB. This antibody is useful as a loading control for Western Blotting due to its high conservation across species and tissues. However, it is essential to note that β-Actin expression levels may vary in certain cell types, such as adipose tissue, and therefore may not be suitable as a loading control in these cases.
Reagents or buffers: RIPA buffer, SDS-PAGE gel, transfer buffer, blocking buffer, and ECL substrate.
Sample preparation: Harvest cells, wash with PBS, and lyse in RIPA buffer. Separate proteins by SDS-PAGE and transfer to a PVDF membrane.
Antibody incubation: Incubate the membrane with the Anti-ACTB Monoclonal Antibody at a suitable dilution in blocking buffer for 1-2 hours at room temperature or overnight at 4°C.
Washing or detection: Wash the membrane with TBST, and detect using an HRP-conjugated secondary antibody and ECL substrate.
Technical notes: The optimal dilution of the antibody should be determined by the user. It is recommended to use a positive control, such as a cell lysate with known ACTB expression, to verify the specificity of the antibody.
Store at -20°C, handle with care, and optimize antibody dilution for each application to ensure specific and sensitive detection.
