The Biotin/HRP antibody is a versatile tool for detecting biotinylated molecules in various applications.
For Western blotting, prepare samples by separating proteins via SDS-PAGE and transferring them to a membrane. Block the membrane with 5% non-fat milk in TBS-T for 1 hour. Incubate the membrane with the Biotin/HRP antibody at a dilution of 1:1000-10000 in TBS-T for 1 hour. Wash the membrane three times with TBS-T, and detect using a suitable HRP substrate.
Technical notes: Optimize the blocking and washing conditions to minimize background noise.
For Immunohistochemistry on paraffin-embedded tissues (IHC-P), deparaffinize and rehydrate tissue sections. Antigen retrieval can be performed using heat-mediated methods. Block endogenous peroxidase activity and incubate sections with the Biotin/HRP antibody at a dilution of 1:100-1000 for 30 minutes to 1 hour. Use a biotin-binding detection system according to the manufacturer's instructions.
Technical notes: Consider using a biotin-blocking step if endogenous biotin is a concern.
For ELISA, coat the plate with biotinylated molecules overnight. Block the plate with 5% non-fat milk in PBS for 1 hour. Incubate the plate with the Biotin/HRP antibody at a dilution of 1:1000-10000 in PBS for 1 hour. Wash the plate three times with PBS-T, and detect using a suitable HRP substrate.
Technical notes: Optimize the coating and blocking conditions to achieve the best signal-to-noise ratio.
Store at 2-8°C. Handle with care to avoid contamination. Use appropriate controls and optimize antibody dilutions for each application to ensure specificity and sensitivity.
